Growth curve and rate of cell division calluses ingazeiro
Abstract
The aim of this study was to describe the growth curve derived callus of leaf explant ingazeiro and observe the rate of cell division during the curve. The leaf explants were inoculated on MS medium, 3% sucrose, 0.7% agar and 9 mM of 2,4-D. At intervals of 10 days, the calluses were weighed. To obtain the mitotic index, the calluses were collected every 20 days, fixed in Carnoy subsequently transferred to 0.5 mL of McIlvaine buffer solution for 10 minutes, sieved and centrifuged. The precipitate was resuspended in McIlvaine buffer and 20μL of this cell suspension were transferred to glass slides and stained with DAPI. The growth curve showed phases: lag (from 0 to 40 days after inoculation), exponential (40th to 50th day), linear (50ºao 70th day) and deceleration (70 to 90ºdia). The rate of cell division followed the curve.
